Melanoma and non-Melanoma Skin Cancer in In ammatory Bowel Disease Patients following Tumor Necrosis Factor-α Inhibitor Monotherapy and in Combination with iopurines: Analysis of the Food and Drug Administration Adverse Event Reporting System

چکیده
Background & Aims: Reports have shown an increased risk of melanoma skin cancer (MSC) with exposure to tumor necrosis factor alpha (TNF-α) inhibitors and non-melanoma skin cancer (NMSC) with thiopurine exposure in in ammatory bowel disease (IBD) patients. Using the Food and Drug Administration Adverse Event Reporting System (FAERS) we sought to evaluate the odds of developing MSC and NMSC for patients on TNF-α inhibitors as monotherapy and in combination therapy with thiopurines and/or steroids. Methods: e FAERS was queried for reports between January 2003 and June 2012. A proportional reporting ratio (PRR) metric analyses was performed on the data to determine the odds of developing MSC and NMSC. Results: e PRR analysis showed increased odds of developing MSC and NMSC for patients on a TNF-α inhibitor (p-value = 0.035 and p-value = 0.03, respectively) and those on a TNF-α inhibitor in combination with a thiopurine (p-value < 0.001 and p-value < 0.001). Conclusion: TNF-α inhibitor monotherapy or use with concomitant thiopurines in patients with IBD is associated with higher odds of developing MSC and NMSC.

Infectious Complications of TNF-α Inhibitor Monotherapy versus Combination erapy with Immunomodulators in In ammatory Bowel Disease: Analysis of the Food and Drug Administration Adverse Event Reporting System*
Background & Aim: Incremental increase in the risk of serious infections with combinations of tumor necrosis factor-alpha (TNF-α) inhibitors and immunomodulators compared to monotherapy with these agents in in ammatory bowel disease (IBD) is unclear. Our aim was to analyze whether there is such an incremental increase in the odds of serious infections. Methods: e FDA Adverse Event Reporting Sys...
متن کاملNon melanoma skin cancers: a retrospective study in department of radiation oncology, Mashhad, Iran
Background: Non melanoma skin cancer (NMSC) is the most common cancer worldwide. In most cases, the general outlook is excellent; however, local recurrence or metastasis can occur. In this study, we investigated possible tumor and host characteristics affecting the treatment outcome in our department of radiation oncology.Method: We retrospectively studied cases with non melanoma skin cancer (N...

Introduction: Due to the high global incidence of skin cancer, its adverse effects on health, and imposition of exorbitant treatment costs on people, the ecological factors associated with non-melanoma skin cancer (NMSC) were examined in patients who referred to the Tumor Clinic of Razi Skin Hospital, Tehran in 2017. Methods: This case-control study was conducted by administration of a researc...
متن کاملAre there Relationships between the VDR-FokI Polymorphism and Vitamin D and the Insulin Resistance in Non-melanoma Skin Cancer (NMSC) Patients? A Protocol for Case-control Studies
Background and Objectives: Evidence indicate relationships between the sunlight exposure, vitamin D status and non-melanoma skin cancer (NMSC) risk. Vitamin D receptor (VDR) polymorphisms have been linked to metabolic changes in insulin resistance and various cancers. The purpose of this study is to investigate relationships between the sunlight exposure and the NMSC risk. Furthermore, relation...
متن کاملInvestigation of TNF-α and IL-6 Levels in the Sera of Non-Melanoma Skin Cancer Patients
Background: TNF-α and IL-6 are both pleiotropic cytokines playing major roles in cancer-associated cytokine networks. They have previously been investigated for their function in skin malignancies, mostly melanomas, and studies on NMSC patients are relatively fewer. In this study, we aimed to investigate the associations of serum levels of IL-6 and TNF-α with NMSCs and its clinicopathological f...
